Effect of Calebin A and/or 5-FU on TNF-β-promoted activation and nuclear translocation of p65-NFκB in CRC cells in monolayer tumor cultures. Serum-starved HCT116 (A) and HCT116R (B) cells in the monolayer culture were treated as described in the Materials and Methods section, labeled for p65 by immunofluorescence and counterstained with DAPI. All experiments were performed at least in triplicate and quantification of positively labeled p65-NF-κB-nuclei and apoptotic nuclei (white arrows) were performed by counting 500–800 cells from 20 different microscopic fields.
