Effects of Ramucirumab and/or Paclitaxel on cell migration and actin polymerization in gastric cancer cell lines. AGS, NCI-N87 [N87], HGC-27 and KATO III human gastric cancer cell lines were cultured for 48 h with 100 µg/ml Ram and/or 1 nM PTX. (a) The results obtained by migration assay performed on fibronectin coated wells is shown on the left. The percentage of migration were calculated at the time T0 and after 48 h (T2). The 100% represented the detection zone completely closed. The experiments were performed in triplicate and the mean values ± SD were plotted in the relative graph. *p < 0.05; **p < 0.001; ***p < 0.0001. The representative images of cell staining with DyLight 554 Phalloidin are presented on the right. Scale bar: 100 µm. (b) The expression of β-tubulin III (TUBB III) and EMT protein E-Cadherin and N-Cadherin after 48 h of Ram and/or PTX treatment was analyzed by Western Blotting in three independent experiments and a representative image is shown on the left. On the right are presented the data of three independent Real Time-PCR experiments of E-Cadherin plotted in the graphs and expressed as mean ± SD. *p < 0.05; ***p < 0.0001.
