A549 cells were incubated with pHLIP constructs or PNA alone at pH 6.2 or pH 7.4 for 2 hours. Confocal microscopy of TAMRA fluorescence (top) and accompanying DIC images (bottom) are shown for the 16-mer pHLIP-PNA construct at pH 6.2 (A) and pH 7.4 (B), the 16-mer PNA alone at pH 6.2 (C), and pHLIP conjugated to TAMRA by a non-cleavable bond (D). Flow cytometry was used to quantify the relative amount of PNA delivered at pH 6.2 vs. pH 7.4 for each pHLIP-PNA construct (E). The respective pH 6.2-to-pH 7.4 delivery ratios are compared in (F).
