Metabolic chemical reporters. (A) Metabolic chemical reporters (MCRs) of protein glycosylation are traditionally per-O-acetylated, azide- or alkyne-bearing analogs of naturally-occurring monosaccharides. Upon treatment of living cells, MCRs can then be metabolized, resulting in protein labeling. (B) The structure of the potential MCRs studied here, 2-azido-2-deoxy-mannose (Ac42AzMan) and 4-azido-4-deoxy-galactose (Ac44AzGal).
