Human macrophage THP-1 cells were seeded into six-well plate and starved overnight. After that, cells were treated with different concentrations of Ang II (0, 0.1, 1 and 10 μM) for 24 h, respectively. After that, cells were harvested to analyze the expression of sST2 (A) and miR-202-3p (B). The mRNA levels were normalized to β-ACTIN mRNA and the levels of miRNA were normalized to U6b. All data were expressed as mean ± SD and the differences of gene expression between groups treated with different doses of ang II were examined by Student’s t test. *P<0.05, **P<0.01, ***P<0.005 vs untreated control. (C) THP-1 cells were seeded into six-well plate and transfected with miR-202-3p mimic or negative control mimic (NC). At 24 h post transfected, cells were further stimulated with 10 μM Ang II for another 24 h. After that, cells were collected to analyze the expression of sST2.
