Breeding of the Atoh1-Cre30 to the CAG-βgeo-tTA-IRES-GFP31 mouse allow for Cre-mediated loxP recombination, removal of a transcriptional 3x poly A (stop) sequence located between the βgeo promoter and the tTA coding sequence, and activation of the tetracycline-controlled transactivator (tTA, Tet-OFF) and downstream GFP reporter. Further breeding of the Atoh1-Cre; CAG-βgeo-tTA-IRES-GFP to the TetO-CycD1-IRES-Luc36 unleashes activation and overexpression of the CycD1 transgene.
