N3-kethoxal (blue star) reacts with single-stranded guanines in the genome (resolved by DNA-binding proteins, such as Pol II as shown in yellow), which can be further biotinylated (red) and enriched for sequencing. The whole process takes 6–7 h in total, from live cell labeling to finish library preparation.
