miR-449a Negatively Regulates Notch1 and Notch2 in LCa Cells
(A) The figure shows representative interaction models between miR-449a and Notch molecules (Notch1 and Notch2). The bindings, predicted by TargetScan, show that Notch1 and Notch2 are putative target genes of miR-449a. (B) The Notch1 and Notch2 mRNA levels were measured in Hep-2 cells transfected with miR-449a mimic or inhibitor, or the corresponding NC using RT-qPCR. HPRT1 mRNA was used as a normalizer. (C) The protein expression levels of Notch1 and Notch2 were determined in Hep-2 cells at 48 h after the transfection of miR-449a mimic or inhibitor, or the corresponding NC by western blot analysis. α-Tubulin was used as a loading control of each western blot (WB). Each sample was run in triplicate. Error bars show mean ± SD. The t test was used for the calculation of p values. ∗p < 0.01, ∗∗p < 0.001.
