(±)-Citalopram-induced inhibition of [3H]imipramine binding to either hα3β4 (□) or hα4β2 (○) AChRs in the desensitized state. Each AChR-containing membrane (1.5 mg/mL) was pre-incubated (30 min) with 15.2 nM [3H]imipramine in the presence of 0.1 μM (±)-epibatidine (receptors are mainly in the desensitized state), and then equilibrated with increasing concentrations of (±)-citalopram. Nonspecific binding was determined at 100 μM imipramine. The plots are combinations of 2-4 experiments, each performed in triplicate, where the error bars are the S.D. The IC50 and nH values were obtained by nonlinear least-squares fit of the plots (r2 = 0.95 for both). The Ki values, calculated using Eq. (1), were summarized in Table 2.
