LIPG knockdown sensitizes to cisplatin and oxidative stress induces the expression of genes involved in inflammation. (A) PEA1 cells were transfected with control siRNAs or LIPG-directed siRNAs. Twenty-four hours after transfection, cells were treated with 20 μM cisplatin for additional 48 h. Subsequently, apoptosis was measured by a luminescent caspase 3/7 activity assay. Data are expressed as mean ± S.E.M. from four independent experiments with technical triplicates each. The numbers above the bars indicate the statistical significance (p-value), based on the two-tailed unpaired Student’s t-test. (B) Real-time RT-PCR analysis of expression of indicated genes in PEA1 cells treated with 20 μM H2O2 for 9 h. All data are expressed as mean ± S.E.M. of delta-delta Ct (ddCt) from four independent experiments with technical triplicates each. Numbers indicate the statistical significance (p-value), based on the Student’s t-test (significant values are highlighted in red).
