The effects of PGD2 and 15d-PGJ2 on the expression of the motor neuron-specific protein marker Islet-1. Undifferentiated NSC-34 cells were treated with vehicle (0.04% DMSO), 15 μM PGD2, or 8 μM 15d-PGJ2 with or without 10 μM GW9662 (a PPARγ antagonist) for 24 h. Equal amounts of cell lysate (10 μg) were analyzed by Western blotting and the Islet-1 antibody and β-actin antibody as an internal control (upper panel). The relative band density was estimated quantitatively using Scion imaging software and is expressed as the ratio of the Islet-1 band intensity relative to the β-actin band intensity. Each value represents the mean ± SD of three different experiments. * p < 0.05, ** p < 0.01, *** p < 0.001.
