Two allele-specific upstream primers that contain the SNP of interest at their 3′ ends and one common downstream primer were used in the amplification reaction. Only the perfectly complementary upstream primer to the target was extended by the DNA polymerase, while the amplicons were detected by the DNA intercalating dye, SYBR Green I.
