The proposal distribution of $s_0^{\text{∗}}$ is a normal distribution restricted to [0, 1] with reflection at the boundaries, denoted here by ${\mathcal{N}}_{\left[\mathrm{0,1}\right]}$. Further, initial $F_0={\text{med}}_{T_0\le t_i<-T}\left\{y_i\right\}$ is the median of the total observed fluorescence in the ROI, taken over all bleaching frames.
