The inhibition of ERK1/2 suppresses morphine-induced releasing of HSP70 and attenuates the ER stress and the phosphorylation of PKA and NR-1. Representative western blot images were shown in this figure. (A) Glibenclamide abolished the phosphorylation of ERK1/2 elevated by morphine. Glibenclamide (200 μM) was given 15 min before morphine (200 μM, 1 h) administration. Cell extracts were collected and analyzed by western blot (n = 3). (B) ERK1/2 inhibitor, SCH772984, could suppress the phosphorylation of ERK1/2 induced by morphine treatment in SH-SY5Y cells. SCH772984 (2 μM) was given 15 min before morphine (200 μM, 1 h) administration. Cells were collected 12 h after morphine treatment and analyzed by western blot (n = 3). (C) HSP70 release induced by morphine was reversed by ERK1/2 inhibitor in SH-SY5Y cells. SCH772984 (2 μM) was given 15 min before morphine (200 μM, 12 h) administration.
