Processing of the data acquired with KB illumination. (a) The plane KB illumination profile with yields frequency artifacts emerging from the nanostructures of the mirrors. (b) By positioning a pinhole with a diameter of 3 µm in the focus of the beam, the profile of the illumination is smoothed. (c) An exemplary projection of murine heart embedded in paraffin. (d) The conventional empty beam corrected projection yield artifacts from small variations in the illumination profile. (e) By creating an artificial flat field using a principal component analysis approach these variations can be reduced. (f) The phase reconstruction was performed by the nonlinear-Tikhonov CTF approach.
