Illustrative EThcD spectra of glycopeptides derived from α-S1-casein on a novel N-glycosylation site. Three examples of the same glycopeptide; DTRNESTQNCVVAEPEK of α-S1-casein (66–82), harboring various glycoforms. The EThcD fragmentation of these glycopeptides resulted in full sequence coverage, and the fragment ion (Y1f) of the amino acid backbone with HexNAc(1)Fuc(1) indicated one core fucose. (A) EThcD fragmentation spectra of the glycopeptide harboring a HexNAc(4)Hex(4)Fuc(1). (B) EThcD fragmentation spectra of the glycopeptide harboring a HexNAc(4)Hex(4)Fuc(1)Sia(1). (C) EThcD fragmentation spectra of the glycopeptide harboring a HexNAc(4)Hex(5)Fuc(2). Lower case c in the peptide sequence indicates a carbamylated cysteine.
