miR-193b forms a negative feedback loop with MYC. (A) Promoter reporter assay and (B) chromatin immunoprecipitation-quantitative PCR assay revealed that MYC could directly bind to the promoter region of miR-193b and suppress the expression of miR-193b. (C) Transfection with a miR-193b mimic decreased MYC expression in F5M2 cells, (D) whereas transfection with a miR-193b inhibitor enhanced MYC expression in F4 cells. (E) Immunofluorescence staining of xenograft tumor tissue also indicated a negative influence of miR-193b on MYC expression. Data are presented as the mean ± SD of at least three independent experiments. *P<0.05. LV, lentivirus; miR-193b, microRNA-193b; NC, negative control.
