Inhibition of A) Ecm1 and B) RNMT–RAM N7‐MTase activity toward probe 6 b by selected substrate analogues and sinefungin. All reaction mixtures contained probe 6 b (2 μm) and either SAM (50 μm), Ecm1 (10 nm) or SAM (20 μm), RNMT–RAM (20 nm) in 50 mm Tris⋅HCl buffer (pH 7.5). The mixtures were incubated at 30 °C in a 96‐well plate. The fluorescence measurements were carried out for excitation at 345 nm and emission at 378 nm by using plate reader. Data are the mean±SD of three independent experiments. The determined IC₅₀ and Hill slope values are shown in Table 3.
