Effects of GSK-3β re-expression or silencing in stable KLHL22-expressing or KLHL22-silenced cell lines in vitro and vivo. We evaluated the expression of β-catenin, LEF, Twist1, Snail1 and E-cadherin via Western blotting. (A, B) The expression levels of β-catenin, LEF, Twist1, and Snail1 were reduced and that of E-cadherin was increased when GSK-3β and Axin-2 was overexpressed in KLHL22-silenced HCT116 and SW480 cells. (C) But the levels of these proteins showed the opposite trend when GSK-3β and Axin-2 was knocked down in KLHL22-overexpressing LOVO cells. GAPDH (cell marker) was used as a loading control.
