VSMC which over express AIF-1 show increased lipid uptake and increased CD36 mRNA abundance. (A) Human VSMC were transfected with 30 MOI of AIF-1 or LacZ adenovirus. After 48 h VSMC were starved in 0.1% FCS for 24 h, then incubated with 5 μg/ml of fluorescently labeled ox-LDL (Dil-ox-LDL) for 2 h. Uptake was quantitated by FACS analysis. (B) Adenoviral transduced human VSMC were treated as in “A”, at which time RNA was extracted and receptor abundance quantitated by quantitative RT-PCR. CD36 mRNA was significantly more abundant at all time points assayed, compared with control VSMC (P < 0.05 for all time points). (C) AdAIF-1 VSMC express more CD36 protein compared with AdLacZ controls. Lysates from unstimulated and ox-LDL stimulated VSMC were immunoblotted with the antibodies shown.
