Table 1.

Kinetic parameters for cleavage of the modified target-guide substrate by E. coli RNase E NTD. The rate of cleavage of 0.1, 0.2, 0.25, 0.3, 0.35, 0.4, 0.45, 0.5, 0.55, 0.6, 0.8, 1 and 2 μM of the modified target-guide RNA substrate by 5 nM E. coli RNase E NTD in the absence of small molecule inhibitors and in the presence of 2 mM AS2, AS4 or 5′S1 was measured. Each experiment was performed in triplicate. Data were fitted as described in Materials and Methods to determine the kinetic parameters, Vmax and Km.
