The GCPII reaction cycle. This cycle depicts important steps/structures throughout the reaction pathway. The suggested reaction mechanism was obtained by QM/MM calculations. It is fully consistent with the crystal structure of the substrate bound in the active site of the E424A mutant of GCPII reported here as well as with the structures of the transition state analogue, and enzyme–product complexes reported previously (19,20).
