Figure 5

Effects of glucocorticoid receptor antagonist on corticosterone or dexamethasone-induced HMGB1 release in primary cultured cortical astrocytes. (A) Astrocytes were pretreated with 0.5 μM mifepristone (mife) for 30 min, and subsequently treated with 1 μM corticosterone for 24 h. The amount of HMGB1 in the media was quantified by Western blotting. Representative blots are shown (HMGB1: 30 kDa). The data are expressed as the mean ± S.E.M. ** p < 0.01 vs. basal and ++ p < 0.01 vs. corticosterone alone. (Tukey’s HSD test; n = 6–10). (B) Astrocytes were pretreated with 0.5 μM mifepristone (mife) for 30 min, and subsequently treated with 100 nM dexamethasone for 24 h. The amount of HMGB1 in the media was quantified by Western blotting. Representative blots are shown (HMGB1: 30 kDa). The data are expressed as the mean ± S.E.M. ** p < 0.01 vs. basal and ++ p < 0.01 vs. dexamethasone alone. (Tukey’s HSD test; n = 5–9).
