Investigation of the cellular response to hypoxia and reoxygenation to the three major hypoxia response programs by Western blot. Grp78 (78 kDa glucose-regulated protein) of the unfolded protein response, HIF-1α (hypoxia-inducible factor 1-alpha) as the master regulator of metabolic adaptation to hypoxia, and the phosphorylation of the p70 S6 kinase of the mTOR (mammalian target of rapamycin) pathway were analyzed. The cells were cultured under standard conditions for 14 days (S), under hypoxia (1% O2) for 14 days (14 d), and after cultivation under hypoxia, followed by reoxygenation (10% O2) for 4 h (14 d 4 h). Three biological replicates were analyzed for each cell line. Grp78 was analyzed together with the proteins shown in Figure 2; therefore, the image for alpha-tubulin is the same as in Figure 2.
