In Vitro Validation of Radiotracer Uptake by cNIS+/GFP+ Canine Myoblasts
(A) Measurement of 99 mTcO− uptake efficiency by canine myoblasts sorted for GFP expression. The experiment was performed with cells under proliferation or after 5 days of differentiation. Sodium perchlorate (NaClO4) was used for specific inhibition of NIS activity. NIS− myoblasts were used as negative control. Values were normalized to 1 × 105 cells. *p < 0.05 and ****p < 0.0001 of 3 independent experiments. (B) Imaging of in vitro cells. Different concentrations of cNIS myoblasts, 1 × 106, 0.5 × 106, 105, and twice 0.5 × 105 imaged after incubation with 185 kBq of 99 mTcO4− imaged in a 12-well plate (upper right).
