Varlitinib inhibits CCA cell proliferation.

Notes: (A and B) KKU-214 and KKU-100 CCA cells were treated with varlitinib for 72 hrs before the growth inhibitory effect was determined by a clonogenic survival assay: colonies which contained >50 cells were counted. (C and D) Varlitinib caused cell cycle arrest in the G1 phase. KKU-214 and KKU-100 were treated with varlitinib for 72 hrs and subjected to propidium iodide staining analysis. (E) G1/S phase cell cycle regulatory proteins cyclin D1 and CDK4 expression levels were determined by Western blot analysis after KKU-214 and KKU-100 cells were exposed to varlitinib for 72 hrs. (F) Expression level of cyclin D1 and CDK4 after normalized with β-actin. Data in (B, D and F) are represented as mean ± SD of three independent experiments. Asterisks indicate statistical significance (p <0.001) compared to the control, analyzed by one-way ANOVA.
