Switchable UniCAR system. The UniCAR system is composed of two separate units: (I) T cells genetically engineered with UniCARs comprising a humanized αE5B9 scFv as extracellular domain and (II) a target module (TM) carrying an antigen-binding domain (eg a single-chain fragment variable (scFv)) that is directed against a specific tumor-associated antigen (TAA) and coupled to the E5B9 peptide epitope (E5B9 tag). UniCAR T cells per se are unable to bind to target cells (OFF switch). The cross-linkage of UniCAR T cells with their targets is mediated by a specific TM resulting in UniCAR T cell activation and tumor cell killing (ON switch). Here, we present novel TMs directed against EGFR.
