A) qRT-PCR was performed on MMTV-CD8-IGF1R, MMTV-Her2-Neu, and p53null frozen tumors or control WT mammary glands using a lineage-associated gene panel. n=2 biological replicates. B) Control and pre-neoplastic mammary glands or C) tumors were dissociated, and FACS analyzed for CD24, CD49f, CD29, and CD61 markers to separate the luminal progenitor (LP)(CD61 high,CD29 low), mature luminal (ML)(CD61 low, CD29 low), and basal/myoepithelial populations(CD29+). D) Tumorspheres from MMTV-CD8-IGF1R tumors were plated into differentiation assays as outlined in materials and methods. Biological replicates n=3, mean+/− s.e.m. Representative of 2 experiments.
