M. leprae genotypes identified in patients and contacts from Bangladesh and the percentage of each subtype are shown. M. leprae DNA was isolated from slit skin smears (SSS) and/or nasal swabs (NS). Genotypes were determined by Whole Genome Sequencing (WGS) or Sanger sequencing according to Monot et al. (2009), Truman et al. (2011). The new subtype 1B-Bangladesh was identified by WGS and primers were then designed for use in Sanger sequencing (Supplementary Table S2). 1D-esxA is 1D subtype containing an A at SNP61425 in the esxA gene, traditionally grouped as 1C (Monot et al., 2009; Truman et al., 2011). This SNP is also found in strains from the genotype 3I and 2E (Figure 2, green). 1* are samples with genotype 1 for which the subtype could not be determined due to DNA concentration limit.
