Prime-boost vs. single shot in non-human primate models of an Ebola virus vaccines. Alternative approaches for vaccination can be used depending on the intended use of a vaccine. (A) In the non-human primate challenge model, a DNA prime rAd boost of GP and NP genes confers protection in a lethal challenge model 8 months and longer after the initial immunization.29 (B) In contrast, a single shot of a rAd vector encoding these genes stimulates a less potent immune response but sufficient immunity for rapid vaccination and may be more useful during an acute outbreak.33
