The reduction of NAD+ by HydAB is not ferredoxin dependent. Purified recombinant HydAB (8.1 μg) was added to assays as denoted by + HydAB. The reaction was initiated with the addition of H2. Clostridial ferredoxin was added at a concentration of 10 μM as indicated by + Fdox. The rate of reduction of NAD+ (ΔA340) after the addition of H2 (0.275 ΔA ⋅ min–1) was similar to the change after the addition of clostridial ferredoxin (0.23 ΔA ⋅ min–1).
