ASO‐mediated reduction of FMR1 and FMRP in FMRP expression neurons results in increased spontaneous activity. (a) Concentration response curves for relative FMR1 expression (first column) and FMRP expression (second column) following a two‐week treatment with varying concentrations of two different ASOs shows a robust, concentration‐dependent knockdown of both FMR1 and FMRP. (b) Quantification of spontaneous activity recorded on MEAs two weeks post‐ASO treatment demonstrates increased activity only in C1_2 cultures in which greater than 95% FMRP knockdown is achieved, as denoted by the bar labeled 4%.
