Two sets of (A) RPA or (B) LAMP complete reactions (n = 4) were prepared in advance. 1ng the target DNA (blue solid line) and water (cyan dotted line) were added into the first set of reactions 10 minutes prior to the start of incubation to simulate a 10-minute delay during sample preparation. Immediately prior to the start of incubation, target DNA (red solid line) or water (magenta dotted line) were added into the second set of reactions to simulate non-delayed samples. (C) Two sets of RPA reactions (n = 4) were prepared in the same way as in Fig 3A except that magnesium acetate was added to each sample immediately prior to the start of incubation.
