Cell lysates were immunoprecipitated with a T7 antibody, and samples of total-cell lysates (TCL) and immunoprecipitates (IP) were analyzed by Western blotting. (A) Fbxo45 interacts with NCad but not with ECad. (B) NCad EC1-2 is necessary for interaction with Fbxo45. (C) NCad EC1-2 is sufficient for interaction with Fbxo45. (D) The SPRY domain of Fbxo45 is necessary for interaction with NCad. All deletion mutations in the SPRY domain disrupt Fbxo45 binding to NCad.
