Spontaneous and induced secretion of various mediators by neutrophils derived from MEFV mutation carriers and patients with other inflammatory diseases. Isolated neutrophils derived from controls and from patients with indicated diseases were cultured without stimulation or were stimulated with 10 ng/ml of lipopolysaccharide (LPS) for 5 h and 1 mM adenosine triphosphate (ATP) for 90 min. Part of the cells was cultured with 5 μg/ml colchicine added at time 0. Concentrations of interleukin (IL)-1β, IL-6, IL-8, IL-10, IL-18, IL-1RA, TNFα, S100A12, proteinase 3, and MPO were quantified by multiplexed bead array assay (D). Data were analyzed by Kruskal–Wallis followed by Dunn's multi-comparison test (*p < 0.05, **p < 0.01, ***p < 0.001). Note that different assay systems were used in Figure 1 compared to Figure 2 explaining potential differences in concentrations.
