ACT001 inactivated NF-κB in murine 4T1 cells. (A) The 4T1 cell culture was supplemented with 30 μM or 60 μM ACT001 for 15, 30, 60, and 180 min. Thereafter, levels of IκBα, phospho-IκBα, NF-κB, and phospho-NF-κB in 4T1 cells were determined through Western blotting. (B) After transfection with small interfering RNA targeting NF-κB, granulocyte-macrophage colony stimulating factor (GM-CSF) levels in the supernatants of 4T1 cell cultures were analyzed via ELISA. *P < 0.05 versus the vehicle control.
