The effect of human GDPD3 overexpression on lipid synthesis in primary hepatocytes from Western diet-fed mice. Ten to twelve weeks after the indicated AAV injection (n = 2 per genotype), primary hepatocytes were isolated from Western diet-fed mice (8–10 weeks of feeding) and incubated with the indicated radiolabels for up to 180 min (n = 3 per time point). Uptake of [14C]palmitic acid (A), formation of [3H]TAG (B), [3H]CE (C), and [3H]PL (D) from [3H]oleic acid, de novo synthesis of [14C]TAG (E), [14C]CE (F), and [14C]PL (G) from [14C]acetic acid, as well as expression of human GDPD3 protein (H), were measured.
