The domain structure and glycosylation sites on PD-L1. PD-L1 is a cell membrane protein with four glycosylation sites (G; brown circles) of asparagine residues (N35, N192, N200, and N219) spanning the IgV-like and IgC-like domains of PD-L1. The numbers represent amino acid residues. The estimated molecular weight of PD-L1 polypeptide is about 33 kDa in a non-glycosylated form (left). Glycosylated PD-L1 consists of about 17 kDa of N-glycan moieties in a range of bands at about 50 kDa on Western blots (right). MW: molecular weight; IgV: immunoglobulin variable; IgC: immunoglobulin constant; TM: transmembrane; ICD: intracellular domain
