Activity of 237-derived CAR T cells against various mouse and human cancer cell lines. (A) Mock, 237-CAR, WE-CAR, TNGK-CAR, LGQ-CAR, or 5E5-CAR transduced T cells were cocultured with Tn antigen expressing murine target cells (Ag104A or ID8 Cosmc KO) or with control cell lines (ACosmc, ID8, or 58−/−), for 24 h at 37 °C, 5% CO2 at various target-to-effector cell ratios. IFN-γ released in the supernatants under each coculture condition was measured by ELISA. Activation of transduced cells was measured in duplicate in two independent experiments. (B) Mock, 237-CAR, WE-CAR, TNGK-CAR, LGQ-CAR, or 5E5-CAR transduced T cells were cocultured with Tn antigen expressing human target cells (Jurkat, Jurkat-MUC1 KO, SKOV3-Cosmc KO, or SKOV3-Cosmc KO-MUC1 KO) or with control cell lines (Jurkat-Cosmc+ or SKOV3), for 24 h at 37 °C, 5% CO2 at various target-to-effector ratios. IFN-γ released in the supernatants under each coculture condition was measured by ELISA. Activation of transduced cells was measured in duplicate in two independent experiments.
