Mtb infection induces lysosomal rewiring in macrophages. Mtb infection alters the cellular lysosomal homeostasis by inducing the biogenesis of lysosomes in infected cells. The lysosomal rewiring is mediated by Mtb surface lipids, predominantly sulfolipid (SL-1), which acts by modulating the mTORC1-TFEB axis. Mutant Mtb (MtbΔpks2) that does not produce SL-1 shows attenuated lysosomal rewiring, reduced lysosomal delivery and enhanced intracellular Mtb survival.
