Screening of small molecule that stabilizes TcCet1. A, melting temperatures (Tm) of TcCet1(18–243 Δ55–75) in the presence of 2 mm compounds were determined by thermal shift assay. Tm of TcCet1(18–243 Δ55–75) with DMSO (control) is indicated by the red dotted line (44.6 °C), and that with ATP is indicated by the blue dotted line (46.0 °C). Four compounds (compounds 466, 695, 860, and 951) that significantly increase the Tm are indicated. B, structures of compound 466 (C13H13NO2: 3,4,6,7-tetrahydroacridine-1,8(2H,5H)-dione), compound 695 (C10H11N2O2: 2-(1H-benzimidazol-1-yl)propanoic acid), compound 860 (C11H12N2O: 3-ethyl-2-methyl-4H-pyrido[1,2-a]pyrimidin-4-one), and compound 951 (C10H14N4O2: 1,3-dimethyl-7-propyl-purine-2,6-dione).
