(A) MCF10A-ras cells were treated with Bay11-7082 (5 μM) for 24 hours in the absence or presence of N-acetyl-L-cysteine (NAC) (5 mM) to measure the Annexin V-FITC/propidium iodide positive cell population by flow cytometry. (B) MCF10A-ras cells were treated with the same IKK inhibitor for 24 hours, and the level of proteolytic cleavage of caspase-3 was determined by Western blot analysis. Actin was used as an equal loading control. (C) MCF10A-ras cells were treated with the IKK inhibitor in the absence or presence of NAC for 24 hours and cytotoxicity was measured by the MTT assay. (D). MCF10A-ras cells were treated with the IKK inhibitor for 12 hours, and the catalytic activity of IKK was determined by the kinase assay as described in Materials and Methods.
