PWRN1 is endogenously competing against hsa-miR-21-5p in GBM cells. (A) The 3’-UTR of wild type (WT) human PWRN1 was shown to include a hsa-miR-21-5p binding domain. Alternatively, the binding domain was mutated (MU). (B) In a dual-luciferase activity assay, HEK293T cells, pre-infected with mimics_21 or mimics_C, were then infected with L/C or L/PWRN1 for 48 h. Relative luciferase activities were measured for co-infected HEK293T cells (* P < 0.05). (C) Between L/C-infected and L/PWRN1-infected LN-229 or U-251 cells, qRT-PCR was applied to compare their hsa-miR-21-5p expression levels (* P < 0.05).
