Reverse effect of hsa-miR-21-5p upregulation on PWRN1-overexpression mediated GBM cell proliferation and migration in vitro. (A) L/PWRN1-infected LN-229 and U-251 cells were further transfected with mimics_21 or mimics_C. Then, qRT-PCR was used to assess theirhsa-miR-21-5p expression levels (* P < 0.05). (B) Double-infectedLN-229 and U-251 cells were assessed by a CCK-8 assay, to compare in vitro proliferation between cells double-infected with L/PWRN1 & mimics_21 and cells double-infected with L/PWRN1 & mimics_C (* P < 0.05). (C) Double-infected LN-229 and U-251 cells were assessed by a 24-well transwell assay. After 24 h, cancer cells migrated onto the bottoms of wells were stained. Representative images were compared between L/PWRN1 & mimics_21- and L/PWRN1 & mimics_C-infected cells. (D) Formigrating GBM cells shown in (C), relative migration was quantitatively measured and compared between L/PWRN1 & mimics_21- and L/PWRN1 & mimics_C-infected cells (* P < 0.05).
