Unique mechanism to be exploited for fungal asthma treatment. Alp1 infiltrates the bronchial mucosa and provokes bronchoconstriction by degrading the extracellular matrix (ECM). This results in increased RhoA activation and Ca2+ signaling to GPCR agonists, leading to ASM hypercontraction and AHR. Inhibitors of Alp1-induced ASM hypercontraction may be identified by high-throughput screening (HTS) to treat and reveal underlying mechanisms responsible for AHR.
