The site directed recombination method. PCR fragments are assembled containing 50% mutated and 50% wild type residues at the selected codons. After that, the fragments are cloned into yeast with the help of homologous overlapping regions. This way, mutations/reversions at the different positions can be studied in a combinatorial display.
