ASO-ψ increases apical membrane conductance. (A) Average Ieq traces were calculated from hBE cells isolated from a CF patient donor homozygous for CFTR c.3718-2477C>T (donor 3) treated with vehicle (grey line), ASO-C (black line) or ASO-ψ (green line). Average (B) apical conductance (Ga) and (C) basolateral conductance (Gb) were calculated by fitting impedance spectra data from the patient epithelial cells to equivalent circuit equations that model epithelial membranes after benzamil, forskolin, VX-770 and GlyH-101 additions. The four data points following the initial compound addition were averaged (±SEM, N = 2; two-way ANOVA; Tukey's multiple comparisons test between treatment groups, *P< 0.05, **P< 0.01, ****P< 0.0001; Sidak's multiple comparisons test within treatment groups, #P< 0.05).
