Heatr9 gapmer transfected cell cultures produced less infectious virus in the culture supernatants as determined by MDCK plaque assay compared to non-targeting control gapmer transfected cell cultures (A). Knockdown of Heatr9 resulted in less induction of Ccl5 at the transcript level (B) as determined by qRT-PCR resulting in less protein (C) in infected culture supernatants. Heatr9 knockdown also resulted in less Ccl4 induction (D). Each dot represents an independent experiment. Lines between data points indicate matching cells from the same experiment. All cells were exposed to the transfection reagent. Non-targeting gapmer indicates transfection with a control gapmer with no specific target (Control Gapmer); Heatr9 gapmer indicates transfection with a Heatr9 targeting specific gapmer (Heatr9 Gapmer). For statistical analysis, paired Student’s t tests were performed. Asterisk (*) indicates a p≤0.05. No indication signifies a nonsignificant result.
