PDCoV nsp5 targets pDCP1A for cleavage. (A) HEK-293T cells cultured in 6-well plates were cotransfected with 0.2 μg of PDCoV nsp5 expression plasmid (pCAGGS-nsp5-HA) or vector along with 2.5 μg of Flag-tagged IFIT1, IFIT2, IFIT3, IFIT5, OASL, OAS1, OAS2, TNFSF10, NMI, ADAR1, USP18, DCP1A, UBE2L6, or GBP1 expression plasmid. After 28 h, cells were lysed and detected by Western blotting with anti-Flag antibody. (B) HEK-293T cells were cotransfected with an N-terminal Flag-tagged pDCP1A and various doses of pCAGGS-nsp5-HA. After 28 h, cells were lysed for Western blotting.
