Identification of the isolated MERS-CoV ChinaGD01 strain. (A) Nested RT-PCR targeting the MERS-CoV RdRp and N genes was performed using primers recommended by the WHO. (B) Vero 81 cells were inoculated with virus-containing culture supernatant for 24 h, and sera from the MERS patient and a healthy donor (negative control) were used to detect viral proteins. (C) NGS of nucleic acid extracted from cell culture described above; clean data were mapped to the ChinaGD01 complete genome sequence with 100% coverage.
